Yes - the rifling does eventually wear out. So do the chambers.
They are measured in terms of rifling width (muzzle gauge) and throat erosion (chamber). It usually takes many thousands of rounds. Much more damage is done by improper cleaning and historically through corrosive primers in ammunition.
